The goal of our research is to better understand the effects of chronic metabolic diseases, such as diabetes, on brain function. Given the high utilization of energy by the brain, we aim to explore the implications of altered metabolism at the behavioral, circuit, cellular and molecular levels. Currently, we use animal models of diabetes to identify the ways in which long-term exposure to hyperglycemia can lead to increased susceptibility to stress and to addiction- or depression- or anxiety-like syndromes.

The Brody Lab studies novel approaches to initiate and increase adaptive immune responses against cancer. These include combinations of checkpoint blockade approaches, discovery of novel tumor immune evasion checkpoints, in situ vaccine strategies using Flt3L to mobilize dendritic cells (DC), cytotoxic therapy to induce immunogenic cell death and load DC with tumor-associated antigens, and DC activators such as TLR and other PRR agonists. A primary focus is on strategies with potential for near-term translation to the clinic, with an emphasis on lymphoid malignancies including lymphoma and CLL.

Our overall goal is to develop knowledge that improves the function and/or quality of life for individuals with spinal cord injury by learning about the mechanisms responsible for muscle wasting and weakness, testing effectiveness of medications that might mitigates these adverse changes, and collaborating with clnical partners to translate these advances to the clinic. Work in the laboratory focuses on three main areas of research: preventing or reversing muscle weakness in persons with disabilities; stimulating neuroplasticity and functional recovery after spinal cord injury; and role(s) of Numb in cell fusion and muscle contraction. Our work relies on a wide range of approaches such as molecular and cellular biology, cell culture models, and animal models. To further translate advances in the laboratory we collaborate with investigators who are working with cutting edge rehabilitation medicine techniques to better understand how these benefit skeletal muscle function.

Dr. Gelb has developed an extensive program in genomics focusing on traits associated with heart malformations. The Gelb lab is studying Noonan and related syndromes, which arise from defects in RAS/MAPK signal transduction. Ongoing studies are using RASopathy induced pluripotent stem cell models and fruit fly models to elaborate disease pathogenesis and for drug discovery. The Gelb group is also studying congenital heart disease as a complex genetic trait, working with the Pediatric Cardiac Genomics Consortium (PCGC). The PCGC is applying various ‘omics’ approaches to discover the causes of these birth defects. In addition, the Gelb group is undertaking functional genomics studies using patient-derived induced pluripotent stem cells.

Research on our lab is focused on understanding the mechanisms by which autism-associated mutations lead to the manifestation of behavioral deficits, with an emphasis on the effect of these mutations on the brain oxytocin system, known as a major modulator of mammalian social behavior, and on brain circuits of social behavior. The goal of our lab is to identify the mechanisms of developmental critical periods to establish (1) perception and (2) cognition relevant to neuro-developmental and psychiatric disorders. The Motivational and Affective Psychopathology (MAP) lab focuses on studying cognitive-affective interactions underlying deficits in motivation, reinforcement learning and inhibitory control in mental health disorders, specifically in substance use disorders, using behavioral, computational and neuroimaging techniques. The goal is to identify neurodevelopmental trajectories of these brain functions that underpin the onset of substance use disorders, changes therein during the chronic substance use and disease maintenance and neuroplasticity during treatment and abstinence from substance use.

The laboratory has two current areas of interest: 1) Overcoming treatment resistance in bladder cancer through immune checkpoint blockade. We have developed a new immune competent mouse model of muscle invasive bladder cancer (MIBC) allowing the evaluation of anti-PD-1 therapy alone and in combination with cisplatin based chemotherapy. Studies are focusing on defining which cell populations are responsive and which are potentially resistant using cell lineage and mutational epitope mapping. 2) Understanding the role of cancer dormancy in treatment resistant prostate cancer. Using genetically engineered mouse (GEM) models we strive to understand to contribution of cellular quiescence and dormancy towards therapeutic resistance and clinical recurrence of castration resistant prostate cancer (CRPC).

Exosomes are naturally secreted nanovesicles that can shuttle fortified or diseased material from the cell of their origin to cells at the vicinity or at a distance. Recently, we have discovered that human stem cells secrete exosomes to mediate repair of a diseased heart by transferring their unique repertoire of miRNAs improving cardiac function and reducing fibrosis. We aim to harness the regenerative and communicative potential of stem cell-derived exosomes for novel therapeutic approaches. Our long-term goal is to artificially engineer these powerful nanovesicles for an off-the-shelf-product to treat patients with cardiovascular diseases.

Our lab’s overall vision is to use genomics to build data-driven, predictive models that improve diagnosis, find new drug-able pathways, and personalize treatment of patients with lung cancer and respiratory diseases. We profile clinical samples from patients using next generation sequencing (NGS) technologies (e.g. single-cell transcriptomic, epigenomic, and spatial data) with the goal of unraveling the underlying regulatory mechanisms in normal lung tissue homeostasis and lung disease. Of particular interest is how the interactions of the lung epithelium with the immune and stromal microenvironment change between normal and diseased individuals.

Dr. Zolla-Pazner’s lab has been pivotal in establishing the central role of antibodies in protection from HIV infection. The Zolla-Pazner lab identified antibodies to the V1V2 region of the HIV envelope glycoprotein gp120 as a correlate of protection in recipients of the only vaccine that has been shown to reduce the rate of HIV infection. Dr. Zolla-Pazner and her research team are leaders in the design and development of candidate HIV vaccines that induce antibodies to these critical regions of the HIV envelope
